A new species of Colletoecema (Rubiaceae) from southern Cameroon with a discussion of relationships among basal Rubioideae Naturalis

Colletoecema magna, a new species from the Ngovayang Massif (southern Cameroon) is described and illustrated. A comparative morphological study illustrates the similar placentation and fruit anatomy of the novelty and Colletoecema dewevrei, the only other species of the genus. Colletoecema magna essentially differs from C. dewevrei by its sessile flowers and fruits, the corolla tube that is densely hairy above the insertion point of the stamens and the anthers that are included. Further characters that separate the novelty are its larger leaves, more condensed inflorescences, and larger fruits. Cloning and sequence analysis of tomato cpDNA fragments: towards developing homologous chloroplast transformation vectors Braz. J. Plant Physiol.

With the view of developing chloroplast transformation vectors based on homologous targeting regions for tomato (Lycopersicon esculentum L.), plastid DNA fragments of tomato cv. IAC-Santa Clara were cloned and analyzed. Isolation and cloning of PstI/SalI chloroplast fragments into the pBlueScript vectors yielded the plasmids pIJB1 and pIJB2 with cpDNA fragments of 8.6 kb and 6.4 kb, respectively. DNA sequencing and computer analyses revealed that the tomato sequences cloned display from 93 to 100 % of identity to the respective fragments in tobacco, which is more pronounced in coding regions. Genetic Relationships of Common Bean (Phaseolus vulgaris L.) Race Chile with Wild Andean and Mesoamerican Germplasm Chilean J. Agric. Res.

The Chilean common bean (Phaseolus vulgaris L.) belongs to the cultivated race Chile and its origin is presumably Andean. The objective of this study was to identify the origin of a group of Chilean accessions based on their genetic relationship with wild material from the Mesoamerican and Andean common bean gene pool. To achieve this objective, universal primers of chloroplast DNA (cpDNA) and mitochondrial DNA (mtDNA) were used to detect polymorphism using Polymerase Chain Reaction - Restriction Fragment Length Polymorphism (PCR-RFLP). Abstract Published protocols for chloroplast and mitochondrial DNA enrichment are based on large samples (more than 5 g) which imply high costs and plenty of sample material. Reduction of tissue sample and reagent quantities is possible and convenient. This work describes an efficient miniprep for chloroplast and mitochondria DNA enrichment and extraction from 500 mg of pericarp samples of Manzano chili fruits (Capsicum pubescens R. & P.). Extraction efficiency from fresh fruit samples was 20 µg DNA g-1 for choloroplasts and 33 µg DNA g-1 for mitochondria. Isolation using the recommended miniprep resulted in costs reduction of 160x for chloroplast and up to 8x for mitochondria.
